  Is the 34401A RS-232 interface enabled?  With the 34401A, only the serial or the GPIB IO can be enabled.   If you are using the serial interface the 34401A will display "RS-232" briefly when it is powered up.  Use the front panel menu keys to switch between RS-232 and GPIB.   Are you using the Agilent IO Libraries, Agilent Connection Expert (ACE) to make the connection?

You need to install ACE in order to communicate with your 34401A.  You set up all of your RS-232 settings in that tool, and it installs the underlying visa/sicl needed for communication with the instrument.
